Lab Test Shows Promising Signs of Tuberculosis Antibiotics in Alleviating Autism Symptoms

Research has revealed some interesting facts about the antibiotics that are used for tuberculosis treatment. The research suggests that the antibiotic has the potential to curb impairments that are associated with ASD (Autism Spectrum Disorder.)

The study further highlights the importance of a drug called “D-Cycloserine” that is responsible for boosting the function of an autism related gene called “PCDH10.” The drug was tested on mice and was found to be successful in improving social impairment symptoms.

Prof. Edward Brodkin, senior author from the University of Pennsylvania, and colleagues reported the findings of the study in the online journal, Biological Psychiatry.

Autism is a developmental disorder that is often characterized by complexities in communication, social, and behavioural skills.

The Center for Disease Control and Prevention has estimated that 1 in every 68 children on average is diagnosed with autism spectrum disorder. It also reports that boys are more prone to autism than girls.

Although the exact reasons behind autism occurrence are unclear, studies have continued to stress the role of genetic factors.

One gene that is most likely to be associated with the autism condition is PCDH10.

Prof. Brodkin stresses that PCDH10 can be expressed in the form of the amygdala—a region of the brain that has a direct link to behavioural and social impairments in autistic individuals. This gene is known to affect the neuronal activity, though its exact influences on social behavior are not yet known.

PCDH10 reduces NMDA glutamate receptor subunit levels

Researchers further studied the role of PCDH10 in hopes of better understanding the various deficits in individuals diagnosed with autism.

The team undertook the initial step of analyzing neurons in the amygdala of mice. For the purposes of the study, the mice were genetically engineered, as scientists had to make sure that they lacked one copy of the PCDH10 gene, thus having its function reduced.

Additionally, the researchers found that the neurons showed a tendency toward reducing NMDA glutamate receptors. This proved to be an indicator for disrupted connectivity in neural circuits.

The mice also displayed social deficits, confirming the role of the PCDH10 drug in social behavior.

The social behaviour of the male rodents was seen to be more affected in comparison to their female counterparts. This seems to have a direct correlation with higher incidences of ASD in male human beings.

Social deficits in mice improved with D-Cycloserine

The researchers then moved on to treat the mice with D-Cycloserine. It is an antibiotic that is prescribed for urinary tract infections and tuberculosis treatment. The team also explains that the antibiotic is known to boost NMDA glutamate receptor functions and can also be used to treat depression.

A noticeable change was observed in the rodents with the help of treatment using the antibiotics.

This not only sheds light on the many contributions of alternative medications to combat social deficits in autism, it also highlights the potential treatments for the impairments.

Nevertheless, Prof. Brodkin stresses the importance of having a deeper knowledge to gain a better understanding before the antibiotic can be advisable as an effective and safe treatment for autism related cases.
